Линейной амплифицированной ДНК секвенирование, л. с. при ПЦР — техника секвенирования нативной двунитчатой ДНК после ее амплификации методом полимеразной цепной реакции. Очищенную ДНК и меченый по 5'-концу секвенирующий праймер смешивают с четырьмя реакционными средами для секвенирования по Сэнгеру и добавляют Taq ДНК-полимеразу (см. Taq-полимераза). Реакционные смеси помещаются в амплификатор и по заданной температурной программе осуществляется многократная денатурация и отжиг матричной ДНК и праймеров. Удлинение праймера Taq-полимеразой идет до тех пор, пока инкорпорированный дидезоксинуклеотидтрифосфат не остановит реакцию. В результате образуется серия амплифицированных фрагментов, которые могут быть разделены электрофоретически (см. Электрофорез) на специальном гелед ля секвенирования. Метод позволяет определять до 500 и более оснований за один цикл, не требует большого количества ДНК.
